sdAb anti-Rabbit IgG (Smart Secondaries®) is a single-domain antibody (sdAb) that binds the Fc region of any rabbit IgG specifically and with high affinity, making this secondary tool ideal for demanding immunolabeling and imaging applications.
Like many other mammals, the immune repertoire of rabbits consists of several immunoglobulins (Ig) isotypes (IgA, IgG, IgE, and IgM). However, compared to other species, rabbits possess only one type of IgG carrying either a lambda or kappa light chain. Historically, rabbit antibodies have been a rich source of polyclonal antibodies, which were employed in various fields of life sciences. They are employed either directly from animal sera or as affinity-purified reagents. In addition, advances in antibody engineering over the last decade enabled the generation of monoclonal rabbit antibodies. Independent of its clonality and irrespectively of its VH domain, our single domain antibodies (sdAbs, nanobodies, VHHs) bind the constant domain (Fc) of any rabbit IgG specifically and with high affinity, making this secondary tool ideal for demanding imaging applications.

DBCO- and Biotin-conjugated sdAbs (1:1 labeling on the C-terminus) are lyophilized from PBS pH 7.4. Unconjugated sdAbs are lyophilized from 5 mM KPi pH 6.0, 300 mM NaCl, 0.5 mM EDTA pH 6.0. Reconstitute with 250 µL pure deionized water for immediate use or with 250 µL sterile 50% glycerol for storage at -80°C. The HRP-conjugated sdAbs are lyophilized from HRP protector buffer and should be reconstituted with 200 µL pure deionized water for storage at 4°C or with 200 µL high purity, sterile 50% glycerol for storage at -80°C. Vials containing lyophilized reagent can be stored at 2-8°C for up to 12 months. After reconstitution, store at -80°C for up to 6 months. Working aliquots can be stored at -20°C for up to 4 weeks (Un-, DBCO-, Biotin-conjugated sdAbs) or at 4°C for up to 6 months (HRP-conjugated sdAbs).
