Direct immunofluorescence labeling of a cultured hippocampus neurons. Myelinated neurons were visualized with FluoTag®-X2 anti-CNPase Atto488 (Cat. No. N4102-At488-L; dilution 1:500; green). Nerve-terminals positive for VGLUT1 were visualized with FluoTag®-X2 anti-VGLUT1 AZDye 568 (Cat. No. N1602-AF568-L; dilution 1:500, red). Nuclei were visulized with DAPI (blue).

FluoTag®-X2 anti-CNPase is derived from an in-house developed single-domain antibody (sdAb) that specifically recognizes a CNPase (2′, 3′-cyclic nucleotide 3′-phosphodiesterase), also known as CNP1. CNPase is one of the most abundant membrane-associated enzymes in the myelin sheath of the vertebrate nervous system. It may also be involved in the RNA metabolism of  myelinating cells of the central nervous system (CNS), the oligodendrocytes.
