Most research monoclonal antibodies are generated in mice. These animals have a rich repertoire of immunoglobulins (Ig) displaying various isotypes (IgA, IgD, IgG, IgE, and IgM). Mice IgGs can be further categorized into five subclasses IgG1, IgG2a, IgG2b, IgG2c, and IgG3. In addition to the subclasses, each IgG molecule can be equipped with either a lambda, or kappa light chain (KLC).

The sdAb anti-Mouse kappa light chain Halo fusion combines a species-specific single-domain antibody (sdAb) binding to all mouse Ig isotypes carrying a kappa light chain with a dehalogenase-based technology. The Halo protein tag sequence is fused to the C-terminus of the sdAb anti-Mouse kappa light chain.

Halo refers to a modified bacterial enzyme, haloalkane dehalogenase, engineered to covalently bind synthetic ligands. These ligands can be conjugated to various functional groups such as fluorophores, biotin, or beads, enabling numerous biochemical, proteomic, and imaging applications. When combined with a fluorescent Halo ligand, the sdAb anti-Mouse kappa light chain Halo fusion can be used in immunofluorescence as a secondary reagent specifically binding to all mouse Ig isotypes carrying a kappa light chain.
