The recombinant heavy-chain anti-SARS-CoV-2 Spike protein S1 antibody is a genetic fusion of the single-domain antibody (sdAb) anti-SARS-CoV-2 Spike protein S1 to a Rabbit IgG Fc domain. This bivalent tool binds the SARS-CoV-2 RBD domain of the S1 protein with high affinity and is ideally suited for ELISA and IHC staining. The fused Rabbit Fc enables researchers to employ conventional secondary detection tools commonly used in automated high-throughput-screening devices. The recombinant heavy-chain antibody (rHcAb) preserves all the biophysical features of the high-affinity binding of the anti-S1 SARS-CoV-2 single-domain antibody and is also capable of neutralizing infections by competing with ACE-2 receptor binding as previously demonstrated by the team of Alejandro Rojas-Fernandez at the Institute of Medicine from the Universidad Austral de Chile. Our anti-S1 SARS-CoV-2 sdAb binds to the S1 protein and is able to neutralize the virus entry by blocking the interaction between RDB and the cell surface-exposed receptor.
SARS-Cov-2 Spike S1 Coronaviruses form trimers with the spike glycoprotein (S) to bind its receptor, mediate membrane fusion, and result in the virus entry. Each spike glycoprotein monomer S is about 180 kDa and contains two subunits, the S1 and S2, mediating attachment and membrane fusion, respectively. The S1 subunit of S protein folds in 2 main domains, the N-terminus domain (NTD) and the receptor-binding domain (RBD), which is responsible for binding a surface protein on mammalian cells.
