PFA-fixed Hela cells stably expressing a TOM70-nfRuby-ALFA fusion protein (nf: non-fluorescent) were stained with FluoTag®-X2 anti-mRuby Atto 488 (N3302-At488; dilution: 1:500) and co-stained with a recombinant antibody (rHcAb) anti-ALFA Human IgG1 Fc-fusion (N1586, dilution 1:1000). The anti-ALFA Human IgG1 Fc-fusion rHcAb was detected using FluoTag®-X4 anti-Human IgG conjugated to AZDye568 (N4704-AF568; dilution: 1:500). N4704_FluoTagX4_Biclonal 2E5+2F3_NanoTag

ELISA plates coated with 0.1 pmol of Human IgG1, IgG2, IgG3 or IgG4 were incubated with a dilution series of the biotinylated sdAb anti-Human IgG two-clone mix (Clones 2E5 + 2F3, mix 1:1). Bound biotinylated sdAbs were decorated with Streptavidin-HRP and detected using TMB reagent. This assay demonstrates the specificity of the two-clone blend, which is available as the fluorescently labeled FluoTag®-X4 anti-Human IgG (Cat. No. N4704). Both clones are also available separately in non-conjugated or non-fluorescent forms (Clone 2E5: Cat. No. N4706, Clone 2F3: Cat. No. N4705).

N4706-Biotin (Clone 2E5)_NanoTag

ELISA plates coated with 0.1 pmol of Human IgG1, IgG2, IgG3 or IgG4 were incubated with a dilution series of biotinylated sdAb anti-Human IgG, clone 2E5 (Cat. No. N4706-Biotin). Bound biotinylated sdAb was decorated with Streptavidin-HRP and detected using TMB reagent. Note that clone 2E5 exhibits the highest affinity for Human IgG1 and IgG4, a lower affinity for Human IgG2 and only weak binding to IgG3.

N4705-Biotin (Clone 2F3)_NanoTag

ELISA plates coated with 0.1 pmol of Human IgG1, IgG2, IgG3 or IgG4 were incubated with a dilution series of biotinylated sdAb anti-Human IgG, clone 2F3 (Cat. No. N4705-Biotin). Bound biotinylated sdAb was decorated with Streptavidin-HRP and detected using TMB reagent. Note that clone 2F3 binds to Human IgG1, IgG3 and IgG4 with similar affinity, but does not bind to IgG2.

FluoTag®-X4 anti-Human IgG is a is a blend of two distinct single-domain antibody (sdAb, nanobody, VHH) clones, each carying two fluorophores. These sdAbs can simultaneously bind to the Fc region of Human IgG. The reagent recognizes all four IgG isotypes but exhibits exceptionally high affinity for Human IgG1 and IgG4 (see Figures). This makes it an ideal secondary tool for high-precision imaging applications involving primary human antibodies of the IgG1 or IgG4 isotype. As part of our Smart Secondaries® range, the FluoTag®-X4 anti-Human IgG is particularly suited for One-step immunofluorescence (One-step IF).
